This funding opportunity announcement (FOA) encourages applications that propose to develop studies that will lead to a broad understanding of the natural history of disorders that already do or could potentially benefit from early identification by newborn screening. A comprehensive understanding of the natural history of a disorder has been identified as a necessary element to facilitate appropriate interventions for infants identified by newborn screening. By defining the sequence and timing of the onset of symptoms and complications of a disorder, a valuable resource will be developed for the field. In addition, for some disorders, specific genotype-phenotype correlations may allow prediction of the clinical course, and for other disorders, identification of modifying genetic, epigenetic, or environmental factors will enhance an understanding of the clinical outcomes for an individual with such a condition. Comprehensive data on natural history will facilitate the field;apos;s ability to: 1) accurately diagnose the disorder; 2) understand the genetic and clinical heterogeneity and phenotypic expression of the disorder; 3) identify underlying mechanisms related to basic defects; 4) potentially prevent, manage, and treat symptoms and complications of the disorder; and 5) provide children and their families with needed support and predictive information about the disorder.

Scientific observations, experiments, and simulations are producing data at rates beyond our capacity to store, analyze, stream, and archive the data in raw form. Of necessity, many research groups have already begun reducing the size of their data sets via techniques such as compression, reduced order models, experiment-specific triggers, filtering, and feature extraction. Once reduced in size, transporting, storing, and analyzing the data is still a considerable challenge a reality that motivates SCs Integrated Research Infrastructure (IRI) program [1] and necessitates further innovation in data-reduction methods. These further efforts should continue to increase the level of mathematical rigor in scientific data reduction to ensure that scientifically-relevant constraints on quantities of interest are satisfied, that methods can be integrated into scientific workflows, and that methods are implemented in a manner that inspires trust that the desired information is preserved. Moreover, as the scientific community continues to drive innovation in artificial intelligence (AI), important opportunities to apply AI methods to the challenges of scientific data reduction and apply data-reduction techniques to enable scientific AI, continue to present themselves [2-4].The drivers for data reduction techniques constitute a broad and diverse set of scientific disciplines that cover every aspect of the DOE scientific mission. An incomplete list includes light sources, accelerators, radio astronomy, cosmology, fusion, climate, materials, combustion, the power grid, and genomics, all of which have either observatories, experimental facilities, or simulation needs that produce unwieldy amounts of raw data. ASCR is interested in algorithms, techniques, and workflows that can reduce the volume of such data, and that have the potential to be broadly applied to more than one application. Applicants who submit a pre-application that focuses on a single science application may be discouraged from submitting a full proposal.Accordingly, a virtual DOE workshop entitled Data Reduction for Science was held in January of 2021, resulting in a brochure [5] detailing four priority research directions (PRDs) identified during the workshop. These PRDs are (1) effective algorithms and tools that can be trusted by scientists for accuracy and efficiency, (2) progressive reduction algorithms that enable data to be prioritized for efficient streaming, (3) algorithms which can preserve information in features and quantities of interest with quantified uncertainty, and (4) mapping techniques to new architectures and use cases. For additional background, see [6-9].The principal focus of this FOA is to support applied mathematics and computer science approaches that address one or more of the identified PRDs. Research proposed may involve methods primarily applicable to high-performance computing, to scientific edge computing, or anywhere scientific data must be collected or processed. Significant innovations will be required in the development of effective paradigms and approaches for realizing the full potential of data reduction for science. Proposed research should not focus only on particular data sets from specific applications, but rather on creating the body of knowledge and understanding that will inform future scientific advances. Consequently, the funding from this FOA is not intended to incrementally extend current research in the area of the proposed project. Rather, the proposed projects must reflect viable strategies toward the potential solution of challenging problems in data reduction for science. It is expected that the proposed projects will significantly benefit from the exploration of innovative ideas or from the development of unconventional approaches. Proposed approaches may include innovative research with one or more key characteristics, such as compression, reduced order models, experiment-specific triggers, filtering, and feature extraction, and may focus on cross-cutting concepts such as artificial intelligence or trust. Preference may be given to pre-applications that include reduction estimates for at least two science applications.

The Office of Planning, Research and Evaluation (OPRE), within the Department of Health and Human Services' (HHS) Administration for Children and Families (ACF), plans to solicit applications for cooperative agreements under the Child Care and Development Fund (CCDF) Lead Agency Data and Research Capacity: Planning Grants (Phase I). These projects are meant to support partnerships between CCDF Lead Agencies and researchers to develop and improve state, territory, and Tribal data systems and build the capacity of CCDF Lead Agencies to collect, analyze, and use data to guide child care policy decisions and program improvement efforts. Sponsored projects will work in collaboration to: (1) assess the current data and research capacity of the CCDF Lead Agency; (2) develop questions of interest to investigate state, territory, and Tribal child care policies and practices; (3) develop logic models to identify the data needed to address the questions of interest; (4) identify data sources available to answer questions of interest and assess the accessibility of those data; (5) identify possible data sources from other state, territory, Tribal, and local data systems for linking; (6) determine whether there is a need to collect data to answer policy-relevant questions; and (7) address the barriers to collecting, analyzing, and using data to inform child care policy decisions. These projects are intended to build the capacity of CCDF Lead Agencies to use data, including data on children, families, the workforce, and providers that participate in the child care subsidy system, to make data-informed decisions to improve child care policies and practices. These 18-month projects, with one project and budget period, will fund a planning phase to develop a research plan to address questions of interest to the CCDF Lead Agency, including a plan for identifying, linking, and using state, territory, and Tribal data to inform child care policy decisions. These planning projects may be followed by a second competition (Phase II), under a separate Notice of Funding Opportunity, to support execution of the research plans to develop and improve state, territory, and Tribal data systems. Projects must be conducted through partnerships between CCDF Lead Agencies (i.e., states, territories, Tribes, or local subsidy administering agencies) and researchers from institutions of higher education, within the state agency, research organizations, and/or other organizations with proven expertise conducting policy research. Applications are invited from CCDF Lead Agencies, institutions of higher education, research organizations, and other organizations with proven expertise conducting policy research. The research supported by this program should be collaborative from start to finish. The CCDF Lead Agency and their research partners must work together to assess the current data and research capacity of the CCDF Lead Agency and develop a feasible plan for identifying, linking, and using data to address questions of relevance to the CCDF Lead Agency. Sponsored projects will be expected to participate in a Consortium that will meet and communicate regularly to identify opportunities for coordination, such as to share information on facilitators and barriers to identifying and using data to inform child care policies and methods for linking data across systems, and to develop collective expertise and resources for the field. The Consortiums collaboration will support research capacity and learning within individual projects and across award recipients. Funding is subject to availability of funds and the best interests of the federal government.

The Unaccompanied Children Lived Experience and Youth Engagement Support Program is forecasted to provide funding to establish, manage, and evaluate a Lived Experience Community Council and Lived Experience Leadership Academy comprised of former unaccompanied children and individuals who sponsored children from ORR care. The recipient shall recruit, identify, orient, and train lived experience experts that reflect the demographic diversity (e.g., age, language, etc.) and varied experiences and circumstances (e.g., pregnant/parenting youth, survivors of human trafficking, etc.) of former unaccompanied children and sponsors. Therefore, prior experience developing programming centering lived experience and youth voice and/or supporting unaccompanied children, sponsors, and reunified families is highly desirable. These lived experience bodies have two objectives. The first is to collect feedback from former unaccompanied children and reunified families about their first-hand experience integrating to the United States. The second is to positively impact the lives of Community Council and Leadership Academy members by building their capacity to serve as leaders within their respective communities. The recipient shall provide the necessary resources, such as personnel, to support the implementation of the lived experience bodies. Task areas will include: a) development of the Community Council implementation plan and Leadership Academy curriculum; b) development of a web-based application; c) selection of eligible candidates to the appropriate lived experience body; d) project management; e) interpretation and translation services for, and during, community meetings; f) secure technology for members to participate in virtual meetings; g) compensate members for their time; h) as separate cohorts, coordinate and implement Washington Weeks for the Community Council and Leadership Academy to meet with government leaders and relevant stakeholders, and i) disseminate to the ORR-funded network feedback obtained through the lived experience bodies' work that centers youth voice and lived experience.

Allergic reactions to drugs and vaccines are a serious public health concern. For the purpose of this notice of funding opportunity (NOFO), allergic reactions include not only IgE-mediated reactions, but other immune-mediated and largely unpredictable drug and vaccine reactions. Based on data from 2013-2014, each year in the United States there are an estimated 200,000 emergency department visits for adverse events related to antibiotics. In children 5 or younger, antibiotics cause more than half (56%) of estimated emergency department visits for adverse drug events and 82% of these visits are due to allergic reactions. Allergic reactions to antibiotics include immediate life-threatening reactions such as anaphylaxis, severe cutaneous reactions including Toxic Epidermal Necrolysis and Drug Reaction with Eosinophilia and Systemic Symptoms, and less severe reactions that still limit further use of the drugs. A label of antibiotic allergy leads to the use of alternative, frequently more expensive antibiotics, often with lower efficacy, which may contribute to antibiotic resistance as well as increased mortality and morbidity in hospitalized patients. While allergic reactions to vaccines are less common, the inability to receive a vaccine due to an allergy is problematic to the individual, and the perceived risk of allergic reactions to vaccines contributes to vaccine hesitancy creating a public health issue. Objectives The objective of this NOFO is to solicit innovative projects to study the mechanisms and management of vaccine or antibiotic drug allergy (research on allergic responses to anti-viral, anti-fungal and anti-parasitic drugs will also be considered). The scope of research into antibiotic or vaccine allergic reactions includes, but is not limited to, the following: IgE-mediated and other mechanisms of immediate allergic antibiotic or vaccine reactions Non-IgE-mediated urticarial reactions to antibiotics or vaccines Delayed-type hypersensitivity reactions to antibiotics or vaccines Severe cutaneous adverse antibiotic or vaccine reactions Biomarkers to identify people at risk for reaction or to confirm reactions to specific antibiotics or vaccines Host factors that may predispose to allergic reactions including, but not limited to, host microbiome, genetics, or inflammatory conditions Mechanisms by which specific infections may increase the risk of an allergic reaction to an antibiotic or vaccine Immunomodulatory approaches to treating or preventing immunologic adverse antibiotic or vaccine reactions A secondary objective of this NOFO is to expand the number of investigators working in the field of vaccine and antibiotic drug allergy. Early-stage investigators are encouraged to apply. UG3/UH3 phase transition and Milestones This funding opportunity is designed as a two-stage cooperative agreement in which Project Scientists from NIAID will work with the investigative team. It will support projects that are organized into a two-year UG3 phase followed by a three-year UH3 phase. The UG3 phase may include pilot, observational, or hypothesis-generating high-risk projects. Preliminary data may be helpful but are not required. The use of electronic health records to identify potential participants for mechanistic studies is allowed. The use of human samples such as those related to clinically indicated and routinely used interventions or tests is encouraged. The UG3 must include milestones to determine the success of the project at the end of this phase. Milestones may be negotiated or re-negotiated after award as this program includes the flexibility to quickly revise milestones and/or aims within the scope of the original peer-reviewed application. Following the completion of the UG3 phase, NIAID staff will review the progress made and make the determination on whether the project will continue to the UH3 phase. NIAID support for the UH3 is contingent upon progress made during the UG3 phase, meeting the milestones, programmatic priorities, the original UG3/UH3 peer review recommendations, and the availability of funds. Some projects might not transition from the UG3 to the UH3 phase. Projects supported by the UH3 phase are required to be hypothesis driven, mechanistic, and extend the work initiated by the UG3 phase. Program Director/Principal Investigator (PD/PI) Meeting Attendance A kick-off meeting and annual meetings will be organized by NIAID to facilitate interactions among recipients to share approaches, data, and methods, with the aim of developing a cadre of dedicated researchers to conduct antibiotic and vaccine allergy and advance mechanistic research. These meetings will convene yearly in the Rockville, Maryland area and will be held in person, though a virtual component will be available to allow broader participation from laboratory members. The PD/PI from each award is expected to attend the kickoff meeting and annual meetings in person. Applications that propose the following topics will be considered non-responsive and will not be reviewed: Adverse drug reactions that are predictable and/or related to pharmacologic properties of the drug such as toxicity or overdose Investigation into drugs that are not antibiotics or vaccines (however research on anti-viral, anti-fungal and anti-parasitic drugs is allowed) Investigations into Guillain-Barré Syndrome or drug-induced autoimmune reactions Investigations of central nervous system and other neurologic adverse responses Investigations into the pathogens rather than host allergic responses to the antibiotics or vaccines Applications that do not propose mechanistic research in the UH3 phase Vaccines that are administered via a route that is not the same route of administration as approved by the FDA Vaccines that are used as an immunotherapeutic (e.g. allergen immunotherapy) HIV/AIDS research
